Poxvirus DNA Replication
Poxviruses, of which vaccinia is the prototype, are complex DNA viruses that display an unusual degree of physical and genetic autonomy from the host cell. The vaccinia genome encodes most, if not all, of the functions required for three temporally regulated phases of transcription and for DNA replication (Moss 1990a,b, 1994; Traktman 1990a,b, 1991). In addition, the 192-kb genome encodes numer...
متن کاملPoxvirus DNA replication.
Poxviruses are large, enveloped viruses that replicate in the cytoplasm and encode proteins for DNA replication and gene expression. Hairpin ends link the two strands of the linear, double-stranded DNA genome. Viral proteins involved in DNA synthesis include a 117-kDa polymerase, a helicase-primase, a uracil DNA glycosylase, a processivity factor, a single-stranded DNA-binding protein, a protei...
متن کاملLipid Membranes in Poxvirus Replication
Poxviruses replicate in the cytoplasm, where they acquire multiple lipoprotein membranes. Although a proposal that the initial membrane arises de novo has not been substantiated, there is no accepted explanation for its formation from cellular membranes. A subsequent membrane-wrapping step involving modified trans-Golgi or endosomal cisternae results in a particle with three membranes. These wr...
متن کاملPoxvirus DNA primase.
Poxviruses are large enveloped viruses that replicate in the cytoplasm of vertebrate or invertebrate cells. At least six virus-encoded proteins are required for synthesis and processing of the double-stranded DNA genome of vaccinia virus, the prototype member of the family. One of these proteins, D5, is an NTPase that contains an N-terminal archaeoeukaryotic primase domain and a C-terminal supe...
متن کاملInhibition of Poxvirus replication by streptovaricin.
A component of streptovaricin complex inhibits the replication of poxvirus. To be effective, the inhibitor must be introduced early in the replication cycle; it appears to inhibit early messenger ribonucleic acid synthesis by viral cores, thus interfering with all subsequent events. Neither of the two major components of the complex, streptovaricin A or C, was the active component.
